pDusk and pDawn - Light controlled fluorescent protein expression
These plasmid were created by my good friends Robert Ohlendorf and Andreas Moglich at Humboldt-Universität in Berlin, Germany. Plasmids come pre transformed in BL21 strains of bacteria.
These plasmids are awesome!!!
When in bacteria pDusk expresses DsRed fluorescent protein in the dark and does not in the light, while pDawn expresses DsRed in the light and does not in the dark. The genes that control light activation are constitutively active so these plasmids can be used in most bacterial strains but work best in BL21 and other overexpression strains! These plasmids are Kanamycin resistant. Make your own gene light activated by cloning it between the 5' NdeI and 3' HindIII restriction sites.

Plasmids come ready to use in E. coli BL21 strain of bacteria, just put in a culture containing Kanamycin. Wait till the culture becomes cloudy before exposing to light. Bacteria take 5-10 hours before visible color appears or disappears.
**Perishables included in this kit have been Freeze Dried. They will be viable up to a month at Room temperature**

I got both pDusk and Ddawn, I'm working on cloning in my own gene of interest to get light controlled expression in bacteria. They take a while for expression to change but there are so many cool possibilities with these guys.
